Temodar (Temazolamide) Oral alkylating agent Starting dose: 150 mg/m2/day for 5 days on a 28-day cycle. Next cycle: Adjust dose according to nadir neutrophil & platelet counts in the previous cycle & at the start of each cycle. Nadir usually occurs around day 22 Patients take temodar for a maximum of 2 years, or until disease progression or intolerable side effects occur. ______________________________________________________________ Dose modification: Based on lowest counts at either Day 22 or Day 29: 1. ANC<1,000 or PLT<50,000 -‡ postpone therapy until ANC>1500 & PLT<100,000. Reduce dose by 50 mg/m2/d for subsequent cycle. 2. ANC 1000-1500 or PLT 50000-100000 -‡ Postpone therapy until ANC>1500 & PLT<100,000. Maintain initial dose. 3. . ANC>1,500 or PLT>100,000 -‡ Increase dose to or maintain dose at 200 mg/m2 X 5 days for subsequent cycle. ______________________________________________________________ Side Effects Myelosuppression Nausea & vomiting – mild to moderate Fatigue – mild to moderate Constipation, Headache, Diarrhea
